Vision by Ageback Covid19 Easy Kit;
Our Kit is a Realtime PCR kit for diagnosing Covid-19.
Working Principle:
1. Step: Sample is isolated with the help of Viral RNA isolation kits. (Suggested kits are indicated in the prospectus, while all kits capable of RNA isolation are suitable)
2. Step: For each type, 2 more than the number of analysis are taken with the help of sterile scissors from the test kit which is preserved at -20 Celcius Strips of Covid 19 N1, Covid 19 N2 and Covid 19 IPC strips are taken and placed in blue, red and green racks respectively.
3. Step: with the help of sterile scissors from the test kit which is preserved at -20 Celcius 2 more than the number of analysis are taken NFW (nuclease free water) are cut and placed in the yellow rack.
4. Step: 8.5 ul of NFW is taken from the yellow rack with the help of the 12-channel pipette and is pipetted in the Covid19 N1 strip. Pipetting is repeated 5-10 time for the solution to be homogenously distributed.
5. Step: 8.5 ul of NFW is taken from the yellow rack with the help of the 12-channel pipette and is pipetted in the Covid19 N2 strip. Pipetting is repeated 5-10 time for the solution to be homogenously distributed.
6. Step: 8.5 ul of NFW is taken from the yellow rack with the help of the 12-channel pipette and is pipetted in the IPC strip. Pipetting is repeated 5-10 time for the solution to be homogenously distributed. After this step, the NFW strip is disposed of.
7. Step: from the test kit which is preserved at -20 Celcius, 2 more than the number of analysis are taken with the help of sterile scissors QPCR Mix solution is cut and and placed in the yellow rack
8. Step: 5 ul of QPCR mix is taken from the yellow rack with the help of the 12-channel pipette and is pipetted in the Covid19 N1 strip. Pipetting is repeated 5-10 time for the solution to be homogenously distributed.
9. Step: 5 ul of QPCR mix is taken from the yellow rack with the help of the 12-channel pipette and is pipetted in the Covid19 N2 strip. Pipetting is repeated 5-10 time for the solution to be homogenously distributed.
10. Step: 5 ul of QPCR mix is taken from the yellow rack with the help of the 12-channel pipette and is pipetted in the IPC strip. Pipetting is repeated 5-10 time for the solution to be homogenously distributed.
11. Step: From the kit which is preserved at -20 celcius, 1 Positive Control Strip is taken and placed in the yellow rack. With a single channel pipette, 5 ul is taken and placed in one of the two extra strips.
12. step: NFW tube preserved at -20 celcius is taken out and 5 ul from this tube is placed in the last tube.
At the end of step 12, the PCR setup is ready.
PCR RUN Setup;
a. In Stage 1, Set to 2 min at 25°C; 1 Rep.
b. In Stage 2, Set to 15 min at 50°C; 1 Rep.
c. In Stage 3, Set to 2 min at 95°C, 1 Rep.
d. In Stage 4, Step 1 set to 3 sec at 95°C.
e. In Stage 4, Step 2 set to 30 sec at 55.0°C.
f. In Stage 4, Reps should be set to 45.
